PRINCIPLE
Rapid Ractopamine Test is a lateral flow immunoassay, using the principle of competitive inhibition to detect ractopamine in swine urine specimens. Colloidal gold conjugated with mouse monoclonal antibody against ractopamine are prior dry-immobilized onto a fiberglass paper located in the sample area, and ractopamine-BSA conjugate is coated in the test line. When the sample is added, it migrates by capillary diffusion through the strip rehydrating the gold conjugate. When present in the urine specimen, the ractopamine competitively prevent the combination of ractopamine-BSA conjugate and gold conjugate, as a result, no colored band appears in the test line zone, indicating a positive result. Whereas, a colored band appears in the test line zone when ractopamine is absent in the urine specimen, which indicates a negative result.

TEST PROCEDURE
(Bring test devices and specimens to the room temperature (15-28° C) before proceeding. )
- Remove the test card from the sealed foil pouch and place it on a flat surface, label the test card with specimen identity.
- Withdraw the urine samples with the pipette in a vertical position, dispense 2~3 drops ( approx 80~120μL ) sample into the sample well marked as "S" on the test card.
- Read the result at 3-5 minutes after adding the sample. Results read after 5 minutes may not be accurate.
INTERPRETATION OF RESULTS
Negative: Both test line(T) and control line (C) appear a colored band.
Positive: One colored band appears in control line zone. No colored band is found in test line zone.
Invalid: If there is no colored band in control line zone of the strip,the test result is invalid. Retest the sample with a new device
