3. Usage :This test kit is used to clinical in vitro detection of N-terminal pro-Brain Natriuretic Peptide (NT-proBNP) concentration in human blood quantitatively.

4. Principle
This test kit use double antibody sandwich method, to detect human NT-proBNP level in sample. The purified
NT-proBNP coated on NC membrane. When testing, put sample into buffer and mix it evenly, there is fluorescein labeled NT-proBNP antibody in buffer. If there is NT-proBNP antibody in sample, it can combine with fluorescein labeled NT-proBNP antibody in buffer, form a complex, affected by chromatography, it reacts with coated NT-proBNP antibody when coming to the place which coated NT-proBNP antibody. If no NT-proBNP antibody in sample, then no reaction. When the reaction over, put the test card into Fluorescence Spectrometer, calculate the NT-proBNP antibody concentration by the strength of the fluorescence signal.

7. Sample requirement
1)This test kit is used to detect hs-CRP concentration in human whole blood,Serum or Plasma quantitatively.
2)Serum: put the whole blood at room temperature for 2 hours or centrifuge at 1000g for 20min after overnight at 4℃, take the supernatant (up-layer) to test, if can not test immediately, store the supernatant at -20℃ or -80℃, avoid repeated freezing and thawing.
3)Plasma: use EDTA or heparin as an anticoagulant, collect sample, centrifuge at 1000g for 15min in 30 minutes, take the supernatant (up-layer) to test, if can not test immediately, store the supernatant at -20℃ or -80℃, avoid repeated freezing and thawing.
4)Whole blood: take venous blood, test immediately; or use EDTA or take heparin as anticoagulant to collect anticoagulant, but should avoid hemolysis.
5) Return sample into room temperature (18~30℃) for 30mins before use.
6) Mix sample to even completely when dilute sample, avoid bubbles, it can mix up-side-down, do not mix it on mixer strongly.
7) Wear mask and gloves when collecting and treating sample, avoid sweat, saliva and aerosol polluting the sample.
8) If sample need transport before use, recommend to store at 2-8℃ (ice bag) when transport. 

8. Test procedure
1) Reagent preparation: Incubate at room temperature (18~30℃) for 15~30min.
2) Sample preparation: Add 90ul serum or plasma (whole blood 150ul) into buffer, mix evenly.
3) Adding sample: add 75ul mixed buffer into sample well accurately, react to 10min.
4) Test: Put the well reacted test card into Fluorescence Spectrometer, the result will show on its screen.

10. The reference value (reference range)
<75 years old: 300pg/mL
>75 years old: 450pg/mL

11. The result interpretation
For NT-proBNP Positive result, it should combine with the patient history, clinical signs and symptoms for a comprehensive judgment. NT-proBNP concentration <300 pg/mL, Negative; 300~50000 pg/mL, recommend dynamic observation; for sample >50 ng/mL, dilute sample accurately and test again. The measured NT-proBNP concentration multiply the dilution factor, the actual concentration of NT-proBNP will be obtained.

12. Limitation
Any negative result from this test kit can not rule out the possibility of heart failure , suggest that the clinical physicians make a comprehensive judgment according to patients' symptoms, medical history, examination and other cardiac biomarkers.    

13. Performance indicators
1). The minimum detection quantity: 100pg/mL.
2). Linear range: 100pg/mL~50000pg/mL, the correlation coefficient r should not be less than 0.98.
3). Stability: store at 37 ℃ for 3 days, the test results should reach the standard.

14. Note
1) Incubate the test kit at room temperature for 15-30mins before use.
2) When use, shake the reagent in bottle evenly firstly, do not mix use reagents from different lot no.
3) Control the reaction time strictly.
4) This test kit is disposable, the used test kit, sample etc. waste should treat as sources of infection according to national relevant regulation.
5) Process as instruction strictly, the result must based on Fluorescence Spectrometer, invalid result need to re-test.
6) There is preservatives in buffer, which has mild toxicity, should avoid contact with clothing, eyes or hands and other exposed parts of the skin.
